- Jeden z tych produktów hybrydyzuje z pojedynczym 3,7-kilozasadowym RNA ograniczonym do śródmózgowia i pnia mózgu szczura, gdzie jest wysoce wzbogacony w serotonergiczny kompleks raphe. <em>Transfekcja</em> pojedynczym komplementarnym klonem DNA pnia mózgu o masie 2,3 kilozasad wystarcza do wywołania ekspresji zależnego od Na(+) <em>5</em>transportera</em>HT na nienerwowym komórki, z transportem selektywnie i silnie antagonizowanym przez antydepresanty specyficzne dla wychwytu <em>5</em>HT, w tym paroksetynę, citalopram i fluoksetynę.
Transfekcja genów oparta na wektorach niewirusowych za pomocą indukowanych ludzkich pluripotencjalnych komórek macierzystych kardiomiocytów.
- Niewirusowa <em>transfekcja</em> ssaczych kardiomiocytów (CM) jest wyzwaniem. Obecne badanie ma na celu scharakteryzowanie i określenie wydajności <em>transfekcji</em> genu opartego na wektorach niewirusowych za pomocą indukowanych ludzkich pluripotencjalnych komórek macierzystych (hiPSC) pochodzących z kardiomiocytów (hiPSC-CM). hiPSC-
- CM odróżnione od PCBC hiPSC zostały wykorzystane jako model komórkowy do transfekcji plazmidami niosącymi białko zielonej fluorescencji (pGFP) przy użyciu polietylenoiminy (PEI), w tym <em>Transporter</em> <em>5</em> <em>Transfekcja </em> <em>Odczynnik</em> (TR<em>5</em>) i PEI2<em>5</em> oraz liposom, w tym lipofektamina-2000 (Lipo2K), lipofektamina-3000 (Lipo3K ) i Lipofectamine STEM (LipoSTEM).

Odkryliśmy, że najwyższą skuteczność <em>transfekcji</em> genu w hiPSC-CMs w 14 dniu skurczu może osiągnąć LipoSTEM, która wyniosła około 32,<em>5</em> ± 6,7%. Spowodowało to jednak również słabą żywotność komórek (60,1 ± 4,<em>5</em>%).

- Na podstawie tego zoptymalizowanego warunku <em>transfekcji</em> genu najwyższa skuteczność <em>transfekcji</em> genu wyniosła <em>5</em><em>5</em>,6 ± 7,8% lub 34,1 ± 4% odpowiednio dla linii hiPSC P1C1 lub DP3 pochodzącej od zdrowego dawcy (P1C1) lub pacjenta z cukrzycą (DP3).
- Żywotność komórek wynosiła odpowiednio 80,8 ± <em>5</em> lub 92,9 ± 2,24% dla P1C1 lub DP3. LipoSTEM jest lepszym niewirusowym wektorem do <em>transfekcji</em> genów hiPSC-CM. Najwyższa skuteczność <em>transfekcji</em> genu pGFP może osiągnąć <em>5</em>0% w przypadku normalnych hiPSC-CM lub >30% w przypadku cukrzycowych hiPSC-CM.
Zależna od receptora N-metylo-D-asparaginianu regulacja transportera glutaminianu pobudzającego nośnika aminokwasu 1.
- Neuronowy <em>transport</em> pobudzający nośnik aminokwasów 1 (EAAC1) jest wzbogacony w regiony perysynaptyczne, gdzie może regulować synaptyczne wydzielanie glutaminianu. - Podobną interakcję zaobserwowano w komórkach glejaka C6 i ludzkich embrionalnych komórkach nerki po ko<em>transfekcji</em> z podjednostkami EAAC1 i receptora NMDA znakowanymi epitopem Myc. Współ<em>transfekcja</em> glejaka C6 z kombinacją podjednostek NR1 i NR2 dramatycznie zwiększyła (około 3-krotnie) ilość Myc-EAAC1, którą można znakować nieprzepuszczalnym przez błony <em>odczynnikiem biotynylującym< /em>. W hodowlach hipokampa krótka (<em>5</em> min), silna (100 mikroM NMDA, 10 mikroM glicyny) aktywacja receptora NMDA zmniejszyła biotynylowany EAAC1 do około <em>5</em>0% poziomów kontrolnych .
- Efekt ten był hamowany przez antagonistę receptora NMDA, wewnątrzkomórkowe lub zewnątrzkomórkowe chelatory wapnia lub hipertoniczną sacharozę. Glutaminian, kwas alfa-amino-3-hydroksy-<em>5</em>-metylo-4-izoksazolopropionowy z cyklotiazydem i tapsigargina naśladowały działanie NMDA. Badania te sugerują, że receptory NMDA oddziałują z EAAC1, ułatwiają ekspresję EAAC1 na powierzchni komórki w warunkach podstawowych i kontrolują internalizację EAAC1 po aktywacji. - Skonstruowaliśmy stabilne linie komórkowe wyrażające <em>transportery</em> dopaminy (DA), norepinefryny (NE) i serotoniny (<em>5</em>-HT) poprzez <em>transfekcję</em> sklonowane cDNA. Komórka rodzicielska LLC-PK1 nie wyraża żadnego z tych <em>transporterów</em> neuroprzekaźników.
- Dlatego aktywność transportu monoamin w każdej z tych linii komórkowych wynika wyłącznie z transfekowanego DNA, co pozwala na porównanie na tym samym tle. Profile hamowania leków dla każdej linii komórkowej są różne i zgodne z oczekiwaniami dla każdego <em>transportera</em>.
- Komórki LLC-NET i LLC-DAT transportowały zarówno NE, jak i DA, a oba typy komórek wykazywały niższą KM dla transportu DA niż dla transportu NE.
- Analiza danych Vmax dla komórek LLC-NET sugeruje, że substrat jest związany z <em>transporterem</em> NE podczas etapu(ów) ograniczania szybkości transportu. Analog kokainy, 2-beta-karbometoksy-3-beta-(4-[12<l]jodofenylo)tropan wiąże się z każdym typem komórek i w każdym przypadku jest wypierany przez substrat transportowy.
- Pomiary wiązania i transportu w równoległych hodowlach komórkowych umożliwiły oszacowanie liczby obrotów dla <em>transporterów</em> noradrenaliny, dopaminy i serotoniny.

Promotor pompy eksportu soli żółciowych człowieka jest transaktywowany przez receptor farnezoidowy X/receptor kwasu żółciowego.
- Pompa wydalnicza soli żółciowych (BSEP, ABCb11) ma kluczowe znaczenie dla zależnego od ATP transportu kwasów żółciowych przez błonę kanalikową hepatocytów i do wytwarzania zależnego od kwasów żółciowych wydzielania żółci. Ostatnie badania wykazały, że ekspresja tego <em>transportera</em> jest wrażliwa na przepływ kwasów żółciowych przez hepatocyt, prawdopodobnie na poziomie transkrypcji genu BSEP.
- Aby określić mechanizmy leżące u podstaw regulacji BSEP przez kwasy żółciowe, sklonowano promotor genu BSEP.
